A tomato (Lycopersicon esculentum Mill.) breeding line (81B416) with' resistance to anthracnose caused by Colletotrichum dematium was crossed to three susceptible genotypes. Parental, F1, F2, and backcross populations were analyzed in the cross with `US28', while parental, F1, and F2 populations were tested in crosses of 81B416 with `US141' and 81B9. Inheritance of resistance was primarily additive, but 3- and 6-factor scaling tests indicated the presence of dominance and epistatic effects. The average broad-sense heritability estimate was 0.57; narrow-sense heritability was estimated at 0.42.

Lesquerella fendleri (Gray) Wats., Brassicaceae, is a potential oilseed crop native to the southwestern U.S. The seed oil contains hydroxy fatty acids, similar to castor. Unique properties of the oil, along with coproducts, allow additional applications that would not be in competition with castor. Plants with vestigial anthers were discovered in a bulk population growing in the greenhouse in 1993. The inheritance of the trait was investigated the following three crop seasons. Crosses were made among sterile and fertile plants and reciprocals among fertile plants. Chi-square results indicate the male sterility trait is expressed by a recessive nuclear gene with cytoplasmic influence restoring fertility. Cytoplasmic male sterile lines can be utilized for development of hybrids. Development of lines without male sterility should lead to higher yields than current bulk populations of lesquerella. Hybrid plants and higher yields will enhance the commercialization potential of this new, alternative crop.

Leptine glycoalkaloids found in certain genotypes of Solanum chacoense, a wild potato relative, are resistance factors against the Colorado potato beetle (CPB). To efficiently introgress CPB resistance through leptine production into the cultivated potato, an understanding of leptine inheritance is important. Analysis of sibs within PI lines revealed a wide segregation for level of leptines. Leptine levels ranged from not detectable to 120 mg/100 g fresh weight among six sibs in a PI line, suggesting leptine production may be controlled by a few major genes. TLC analysis of F2 and backcross progeny, however, indicated that several genes probably control leptine level. This apparent multigene control of leptine level may make it difficult to incorporate leptine synthesis into the cultivated potato. Therefore, we are presently identifying microsatellite and RAPD markers associated with leptine synthesis to enable marker-assisted selection and facilitate the incorporation of leptine synthesis into the cultivated potato.

Starch gel electrophoresis was used to screen 10 enzyme systems for variation in fountain grass, Pennisetum alopecuroides (L.) Spreng. plants exhibiting four different growth habits: dwarf(d), mound(m), prostrate(p), and upright (u). Only phosphoglucoisomerase (PGI; E.C. 5.3.1.9) was found to be polymorphic at one locus, PGI-2, and was expressed as two alleles, which appeared to be associated with growth habit. The dwarf form expressed one slow band (SS), the mound and prostrate forms exhibited one fast band (FF), and the upright form carried triple bands indicating a heterodimer (FS). Hybrids between FF and SS parents were detected as triple bands (FS). Three generations of progeny resulting from 16 crosses and selfs of these growth habits all followed the expected segregation ratios for typical Mendelian inheritance of this isozyme.

The inheritance of two new induced mutations for spindly branch was investigated in common bean (Phaseolus vulgaris L.). Each mutant was found to be controlled by a recessive gene. Allelism tests were performed beween a previously reported spindly branch mutant (sb) and the two new spindly branch mutants; the new mutants were found to be nonallelic to sb and to each other. The gene symbols sb-2 and sb-3 are proposed for the new mutants. Repulsion phase F2 linkage tests were made for all nine combinations of reclining foliage (rf) and sb among the two mimic mutant series rf, rf-2, rf-3 and sb, sb-2, sb-3. No linkages were detected.

The inheritance of an induced mutant for spindly branch and male sterility (SBMS) was investigated in common bean (Phaseolus vulgaris L.) in F2 and backcross populations. The results support the hypothesis that the mutant is controlled by a single recessive gene. Extensive breeding work with SBMS, involving several thousand F2 progeny, produced no recombinant of the types expected if two closely linked genes controlled the character. Therefore, a single pleiotropic gene apparently controls SBMS. Allelism tests demonstrated that SBMS is allelic with sb but not with sb-2 and sb-3. The gene symbol sb ms is proposed for SBMS because it is a new allele at sb, with the order of dominance being Sb > sb > sb ms. Various ways to exploit the new mutant for marked male sterility are discussed.

Prostrate growth habit (PGH) in tomato (Lycopersicon esculentum Mill.) lines derived from breeding material developed at the Agriculture Canada Research Station, Beaverlodge, Alberta, was the subject of a quantitative inheritance study. Plants with PGH have an increased lateral branch angle, relative to upright plants, and crown-set fruit supported above the soil surface making hand harvest easier. Genetic parameters were estimated in two families (20G and 53G), each containing PGH and upright-habit parental lines, F1, F2, and backcrosses to each parent. Field-grown plants were subjectively rated twice during the growing season. Broad-sense heritability of PGH in family 20G was estimated to be 0.65 and 0.71 for ratings of plant growth habit 6 and 9 weeks after transplanting, respectively, and 0.71 and 0.68 for those of family 53G. Narrow-sense heritability was estimated to be 0.83 and 1.05 for the two ratings in the 20G family and 0.77 and 0.78 in the 53G family. F1 and F2 means were not different from mid-parent values. The genetic variance was entirely additive and expression was influenced by the environment. The data did not support the hypothesis that PGH was controlled by a single gene.

Colorado Potato Beetle (Leptinotarsa decemlineata Say., CPB) is a destructive pest of the cultivated potato, Solanum tuberosum L. Certain glycoalkaloids in potato leaves are effective deterrents to this insect; however, in tubers these compounds can be toxic to humans. Leptines are foliar-specific glycoalkaloids produced by the related species, S. chacoense. These compounds have been shown to confer resistance to CPB. We are studying the inheritance of leptine production in segregating F1 and F2 populations derived from two S. chacoense accessions, 55-1 and 55-3, which are (respectively) high and low leptine producers. The F1 segregates 1:1 for high (>70% of total glycoalkaloids) and low (<20% of TGA) leptine content. Segregation data from the F1 and F2 populations suggest a twogene model for leptine production: a dominant repressor and a recessive inducer. Using two bulked DNA samples composed of highand low-leptine individuals from the F1 population, we are using various types of molecular markers (RAPDs, SSRs, DS-PCR, and AFLPs) to search for markers linked to leptine production. We have identified a RAPD band that appears to be closely associated with low leptine content and supports the two-gene model. The use of such a marker in a breeding program will facilitate the development of CPB resistant potato varieties.

Lesquerella fendleri (Gray) Wats. (lesquerella, Brassicaceae), native to the southwestern United States, is a potentially useful industrial oilseed crop. The seed oil contains hydroxy fatty acids, similar to castor (Ricinus communis L.) seed oil. The unique properties of the oil, along with coproducts, allow additional applications that would not compete with castor oil. Plants with vestigial anthers (male-sterile) were discovered in a greenhouse-grown, nonselected population in 1993. The inheritance of the trait was investigated through four crop seasons. Crosses were made among male-sterile and male-fertile plants from an open pollinated population, thus, they were heterozygous for many traits. Statistical analysis indicated that male sterility is expressed as a result of two nonlinked nuclear genes with epistatic relations and different cytoplasms, which cause partial or total fertility restoration. These ratios fit a 13:3 epistatic ratio, indicating that male sterility is controlled by homozygous recessive alleles at one locus in combination with at least one dominant allele at the second locus, i.e., ms1ms1 Ms2_. Some cross results were skewed in favor of fertile phenotypes presumably due to cytoplasmic effects causing partial fertility restoration. Male-sterile lines could be used for hybrid development and this information will be helpful in implementing a strategy for hybrid development. Hybrid plants and higher yields will enhance the potential for commercialization of this new alternative crop.
