Pteroceltis tatarinowii Maxim, the only species of the genus Pteroceltis (family Ulmaceae), is an endemic rare tree species in China. This study was performed to explore vegetative propagation techniques for P. tatarinowii using stem cuttings. First, the effects of exogenous indole-3-butyric acid (IBA) and cutting positions on rooting performance were investigated to screen the appropriate exogenous auxin treatment and to determine the proper cuttings type. The results showed that the control cuttings pretreated with no exogenous IBA, irrespective of whether the stem cuttings were terminal, middle, or basal, rooted in a manner significantly inferior to that of cuttings pretreated with IBA. Their rooting percentage was less than 50%. Among the IBA-treated cuttings, the middle cuttings pretreated with 1000 mg·L−1 IBA rooted best, with the shortest number of days until rooting emergence (20 days), the highest rooting percentage (84.0%), the lowest mortality rate (4.0%), the greatest root number (average of 6.7 per cutting), and the longest roots per cutting (44.4 cm per cutting). Terminal cuttings pretreated with 1000 mg·L−1 IBA acquired satisfactory rooting traits and had the same shortest rooting duration (20 days) and the following parameters: rooting, 70.7%; mortality, 10.7%; average roots per cutting, 5.2; and longest root, 29.1 cm. To further determine the optimum cutting propagation time for this plant, a second experiment was performed and the cuttings were collected beginning in early June, when the growth of the current season was feasible for harvesting cuttings. Stem cuttings collected in late June and middle July had significantly higher rooting percentages (≥80%) compared with those collected in early June (66.7%). The other three rooting parameters were not significantly affected by the collection date. However, according to the overall rooting traits, the cuttings collected in both late June and middle July remarkably outperformed those collected in early June regarding the number of roots and the total root length per cutting. The initial nutrient reserves in the cuttings were also determined. A significant difference in the soluble carbohydrate level was found among collection times, but the nitrogen level in the cuttings was similar. The study revealed that stem cutting propagation of P. tatarinowii was achievable, and it was best achieved with cuttings collected from the terminal and middle positions of the branches of the current season from late June to middle July and treated with 1000 mg·L−1 IBA using the quick dip method.

Acer truncatum seeds are an excellent source of beneficial natural compounds, including high levels of unsaturated fatty acids (UFAs), that promote health. Recently, A. truncatum has emerged as an oil crop. Therefore, the transcriptomes of A. truncatum seeds at 70, 85, 100, 115, 145, 180 days after flowering (DAF) were analyzed to gain a better understanding of the transcriptional and translational regulation of seed development and oil biosynthesis. A total of 28,438 genes were identified, and 3069/2636, 3288/3438, 1319/2750, and 5724/5815 upregulated/downregulated genes were identified when comparing different samples with 85 DAF seeds. Sixteen lipid metabolism pathways with 754 differentially expressed genes (DEGs) were identified, including 34 DEGs associated with UFA biosynthesis. A phylogenetic analysis revealed that six putative fatty acid desaturase (FAD) genes clustered into five FAD groups. A quantitative real-time polymerase chain reaction analysis indicated that the temporal expression patterns of oil biosynthesis genes and transcription factors were largely similar to the RNA sequencing results. The results of this study will enhance the current understanding of oil metabolism in A. truncatum seeds and allow new methods of improving oil quality and seed yield in the future.

The pathogenic fungus Colletotrichum musae infects developing green bananas (Musa spp. AAA group), but remains latent until the fruit ripens. The aim of this research was to determine whether the appearance of disease symptoms is regulated by chitinase gene expression following treatment of fruit with benzothiadiazole (BTH) and methyl jasmonate (MeJA), and with physical (heat) and chemical (H2O2 and Ca2+-related) treatments. In bananas inoculated with C. musae, BTH and MeJA lowered disease severity and stimulated higher gene expression compared with the untreated controls during ripening. However, in naturally infected bananas, BTH and MeJA treatments slightly reduced transcription of the chitinase gene in green bananas, but they prolonged gene expression in ripe bananas and significantly reduced disease severity. The combination of H2O2 and the NADPH oxidase inhibitor, diphenylene iodonium, down-regulated chitinase gene expression and compromised disease resistance compared with H2O2 alone. Heat treatment (HT) or the combination of HT followed by CaCl2 reduced disease, but only the latter significantly upregulated chitinase gene expression. The combination of HT and a calcium ionophore (A23187) resulted in different disease indicies and different levels of gene expression depending upon the order of application: HT followed by A23187 induced higher gene expression and lower disease. The results suggest that disease resistance of green bananas could be related to high and prolonged levels of chitinase gene expression, and chitinase could be involved in harvested banana's anthracnose resistance activated by different defense pathway signals, such as BTH, MeJA, H2O2, and Ca2+.

This study aimed to clarify the relationship between grape (Vitis vinifera L.) quality and environmental factors (climate and soil), and to elucidate the theoretical basis and provide technical guidance for the rational planning of the cultivation area and the precise regulation of the cultivation mode in the future. The fruits of three different grape cultivars, Pinot Noir, Merlot, and Vidal, as well as soil samples and meteorological data were collected from three wine grape growing areas (Wuwei, Zhangye, and Jiayuguan) in the Hexi Corridor, Northwest China. Principal-component analysis (PCA) and correlation analysis were conducted to understand the relationship of grape quality with weather conditions and soil properties. The results showed that the titratable acid content of grape berries was significantly negatively correlated with average temperature from August to September, average minimum temperatures from August to September, and from April to October, mean annual minimum temperature, growing degree days from August to September, and soil total nitrogen content, and significantly positively correlated with average rainfall from April to October, annual mean rainfall, and soil available potassium content. In addition, the pH of grape juice was significantly negatively correlated with annual mean rainfall and soil available potassium content. However, it was significantly positively correlated with average temperature and average minimum temperature from August to September, average minimum temperature from April to October, growing degree days from August to September, as well as soil total nitrogen content. In addition, the results of PCA showed that the fruit quality scores of ‘Vidal’ in Jiayuguan, ‘Merlot’ in Zhangye, and ‘Pinot Noir’ in Wuwei were the highest, respectively. In conclusion, the contents of titratable acid, pH, and Brix° are greatly affected by climate and soil factors. ‘Vidal’, ‘Merlot’, and ‘Pinot Noir’ were suitable for planting in Jiayuguan, Zhangye, and Wuwei, respectively. Nowadays, few reports focus on the relationship between grape quality and soil and climate conditions. Based on the investigation and analysis of the differences of climate, soil, and grape berries quality in the three production areas of Hexi Corridor, the relationship among climate, soil, and grape quality by using statistical methods was studied, which could provide references for clarifying the reasons why environmental factors affect grape quality and select the suitable area for wine grape cultivation.
