Cuttings of Euphorbia pulcherrima Willd. ex Klotzsch `Dark Red Annette Hegg' were grown hydroponically in minus S Hoagland's solution modified to supply 0, 1, 2, 4, or 8 mg S/liter for 8 weeks. Nutrient solution changes; visual observations, sampling of tissue, and measurement of electrical conductivity and pH were done every 2 weeks. Deficiency symptoms appeared after 4 weeks of growth in treatments supplying 0 or 1 mg S/liter and occasionally in treatments supplying 2 mg S/liter. Symptoms included reddening of the petiole and main vein of new leaves followed by yellowing of these leaves. Leaf tissue S levels ranged from 700 to 3600 mg S/kg of plant. Deficient levels were identified as <2200 mg S/kg of plant. Suggested critical tissue levels of S would be 2300 to 3000 mg S/kg of plant leaf tissue.

Imazapic is an acetolactate synthase-inhibiting herbicide labeled for weed control in pastures, rangeland, and noncrop areas. Field research was conducted in Knoxville, TN, USA, during 2020 and 2021 to evaluate the tolerance of four hybrid bermudagrass (Cynodon dactylon × Cynodon transvaalensis Burtt Davy) cultivars to applications of imazapic for growth suppression. Separate experiments were conducted on ‘TifTuf’, ‘Tifway’, ‘Tahoma 31’, and ‘Latitude 36’ hybrid bermudagrass. Experiments included plots (1.5 m2) arranged in a randomized complete block design with four replications and were repeated. Treatments were applied 14 Aug 2020 and 6 Aug 2021, and were mixed with methylated seed oil. Imazapic rates were 0, 35, 52.5, 70, or 105 g⋅ha–1. Cultivar tolerance was assessed via visual ratings of turfgrass injury relative to untreated check plots. Normalized differential vegetation index data were collected on each date turfgrass injury was evaluated. Growth suppression was quantified via reductions in dry clipping weight after mowing. Hybrid bermudagrass injury increased with imazapic rate for all cultivars, and peak injury (> 30%) following all imazapic treatments occurred within 14 days. At the lowest imazapic rate (35 g⋅ha–1), injury was transient, with all hybrid bermudagrass cultivars fully recovered by 28 days. All rates of imazapic reduced hybrid bermudagrass dry clipping weight for 21 days for all cultivars. Further research is warranted to explore lower application rates than those tested in our study, in addition to determining tolerance and growth suppression of other turfgrass species commonly managed on golf courses.

Papaya ringspot virus (PRSV) is a devastating disease that has a detrimental impact on both commercial papaya production and Caricaceae germplasm conservation. In 1998, the PRSV coat protein transgenic line 55-1 and derived progeny were released to growers in Hawaii. The transgenic varieties have provided durable and practical control of the disease that have saved the papaya industry. However, like with transgenic crops throughout the world, there is public concern about the possibility of cross-contamination of these transgenic materials into nontransgenic lines. As the designated germplasm repository for Caricaceae, we are responsible for maintaining the genetic integrity of each accession. Therefore, we have developed a protocol using polymerase chain reaction for detection of the adventitious presence of the 55-1 transgene insertion event in both parental plants and their progeny seed populations. This protocol assures a 99.9% confidence level of obtaining seeds that are 99.5% transgene-free. The protocol developed in this study is not typical for most seed validation techniques because there is a higher than normal producer risk resulting from the potential of large numbers of seeds not meeting the stringent criteria. However, we believe this is necessary to ensure the genetic integrity of seeds stored in the repository.
