Dendrobium officinale Kimura et Migo is a famous traditional Chinese medicinal plant. It produces various phytochemicals, particularly polysaccharides, which have nutraceutical and pharmaceutical values. To increase its biomass production and polysaccharide content, our breeding program has generated a series of polyploid cultivars through colchicine treatment of protocorm-like bodies (PLBs). The present study compared two tetraploid cultivars, 201-1-T1 and 201-1-T2, with their diploid parental cultivar, 201-1, in an established in vitro culture system. Tetraploid ‘201-1-T1’ and ‘201-1-T2’ had shorter leaves and shorter and thicker stems and roots, and they produced higher biomass compared with the diploid cultivar. The length and width of stomata significantly increased, but stomatal density decreased in tetraploid cultivars. The PLB induction rates from the stem node explants of the tetraploid cultivars were significantly higher than those of diploid. However, the PLB proliferation of tetraploids was lower than that of the diploid. The mean number of plantlets regenerated from tetraploid PLBs was also lower than that of the diploid after 4 months of culture. Polysaccharide contents in stems, leaves, and roots of 6-month-old tetraploid plantlets were significantly higher than those of diploids. The polysaccharide content in the stem of ‘201-1-T1’ was 12.70%, which was a 2-fold increase compared with the diploid cultivar. Our results showed that chromosome doubling could be a viable way of improving D. officinale in biomass and polysaccharide production.

Seashore paspalum (Paspalum vaginatum) is an important warm-season turfgrass distributed in tropical and coastal areas. It has excellent resistance to abiotic stresses, such as salinity, drought, and low temperature. However, the research on genetic diversity of local P. vaginatum collections from China is limited. In this study, the genetic diversity among 58 P. vaginatum accessions from four different provinces in China and four cultivars were assessed using simple sequence repeat (SSR) markers. The results indicated that a total of 45 alleles were detected by 19 polymorphic markers, with a range of 2 to 4 and an average of 2.4 alleles per marker. The genetic similarity coefficients between each pair of the 58 P. vaginatum accessions and four cultivars ranged from 0.51 to 1.00, with an average of 0.77. The range of variation of Shannon diversity index of each SSR marker was 0.047 to 1.075, with an average of 0.486. The polymorphic information content of each SSR marker varies from 0.016 to 0.577, with an average of 0.249. The results of cluster analysis and principal component analysis (PCA) showed that 58 P. vaginatum accessions and four cultivars were divided into four groups. These results provide the theoretical basis for the genetic diversity assessments and molecular marker–assisted breeding of P. vaginatum species.

Chinese cymbidiums are important flowering ornamental plants. Traditional propagation via seed or division cannot satisfy growers’ demand for commercialization of new cultivars, and in vitro propagation has a low micropropagation efficiency due to the browning of rhizomes. In this study, rhizomes of Cymbidium ‘14-16-13’ and ‘14-16-5’ were cultured on half-strength Murashige and Skoog (MS) medium supplemented with 6-benzyl aminopurine (BAP), NAA (α-napthaleneacetic acid), or BAP with NAA under either the dark or light. The degree of browning was read, and rhizome proliferation or sprouting (sprout numbers) was evaluated. Results showed that there was significant difference in browning grade of rhizomes between ‘14-16-13’ and ‘14-16-5’ regardless of dark and light culture. Dark culture induced rhizome proliferation but failed to induce sprouts. Light culture slightly elevated the degree of browning but induced sprouting. Among the growth regulators evaluated, BAP was more effective for sprout induction. As rhizome browning appeared to be inevitable in micropropagation of the cymbidiums, a compromise between browning and sprout production could be a realistic approach. Our study showed that rhizomes cultured on half-strength MS medium supplemented with 1.5 mg·L−1 BAP were able to produce more than 16 sprouts per vessel even though browning occurred in the rhizomes. Thus, culturing rhizomes in this medium could be a practical solution for in vitro propagation of Chinese cymbidiums.

To select resistant germplasm resources and understand the growth and physiological responses of kiwifruit (Actinidia sp.) to drought stress, five species, Actinidia macrosperma (Acma), Actinidia longicarpa (Aclo), Actinidia deliciosa (Acde), Actinidia hemsleyana (Ache), and Actinidia valvata (Acva), were assessed under tissue culture conditions. Rootless seedlings of five species were cultured in a medium containing polyethylene glycol [PEG (formula weight 8000)] to induce drought stress (0%, 5%, 10%, 15%, and 20%). After a 30-day culture, three growth indices [fresh weight (FW), plant height (PLH), and leaf number (LN)] and six physiological indices were determined, and the drought damage index (DDI) was determined. The DDIs of five species increased, and three growth indices decreased with increasing PEG concentrations. The following changes were observed under 20% PEG treatment conditions: superoxide dismutase (SOD) activities increased significantly in Acma, Aclo, and Ache specimens; peroxidase (POX) activities remained stable in Acde, Ache, and Acva specimens; and catalase (CAT) activities increased sharply in Acma and Acva. Furthermore, the results indicated that soluble sugar (SS) content increased slightly in Acma, Aclo, Acde, and Ache but it decreased in Acva specimens. Proline (PRO) content increased significantly in Acma and Acva, and malondialdehyde (MDA) contents tended to increase under drought stress in all five species. Principal component analysis (PCA) results indicated that the order of drought tolerance in the five genotypes examined in this study under tissue culture conditions was as follows: Acma > Acva > Acde > Aclo > Ache. Therefore, we concluded that Acma and Acva are more resilient germplasm resources that represent promising kiwifruit-breeding materials. Furthermore, tolerance to drought stress in these species should be further investigated under orchard conditions.

The banana, a typical climacteric fruit, undergoes a postharvest ripening process followed by a burst in ethylene production that signals the beginning of the climacteric period. Postharvest ripening plays an important role in improving the quality of the fruit as well as limiting its shelf life. To investigate the role of glutamate decarboxylase (GAD) in climacteric ethylene biosynthesis and fruit ripening in postharvest banana, a GAD gene was isolated from banana, designated MuGAD. Coincidently with climacteric ethylene production, MuGAD expression as well as the expression of the genes encoding the Musa 1-aminocyclopropane-1-carboxylate synthase (MaACS1) and Musa 1-aminocyclopropane-1-carboxylate oxidase (MaACO1) greatly increased during natural ripening and in ethylene-treated banana. Moreover, ethylene biosynthesis, ripening progress, and MuGAD, MaACS1, and MaACO1 expression were enhanced by exogenous ethylene application and inhibited by 1-methylcyclopropene (1-MCP). Taken together, our results suggested that MuGAD is involved in the fruit ripening process in postharvest banana.

Ten polymorphic microsatellite loci were isolated and characterized from an enriched genomic library of Paphiopedilum concolor (Batem.) Pfitzer. The number of alleles per microsatellite locus ranged from three to 11 with an average of 6.4 in a sample of 30 individuals from three populations. The observed and expected heterozygosity ranged from 0.200 to 0.800 and from 0.544 to 0.827, respectively. These microsatellites can be used as tools to investigate the genetic structure of P. concolor populations and relationship patterns with closely related taxa.
